Flow TriCEPS
combined with siRNA knock down
Step 1: Optimization
Binding of the ligand of interest to the cells expressing the unkown targets and off-targets is shown by Flow-TriCEPS using flow cytometry. Further, the binding conditions are optimized (time, temperature, pH, co-factors, cell activation etc.)
Step 2: Identification
The optimized binding conditions are used to identify the targets and off-targets using the LRC-TriCEPS or the Hatric-LRC platforms.
Step 3: Confirmation
The identified target(s) and off-target(s) are knocked down in the original cells using siRNA and the binding of the ligand to the cells is visualized using Flow-TriCEPS. If the binding is reduced compared to the wild type cells, the target / off-target is confirmed.

In a first step binding of your ligand of interest to cells expressing the unknown targets is established in a pretest using Flow-TriCEPS or TAMRA-TriCEPS. In a next step the LRC-TriCEPS or Hatric-LRC experiment is performed to identify the unknown targets of your ligand of interest. We expect a low number of target candidates (less than 5). Now the identified target proteins can be down regulated using siRNA and with Flow-TriCEPS or Tamra-TriCEPS reduced binding is expected of your ligand of interest compared to the wild type cells when the true target is knocked down.

Using LRC-TriCEPS, we aimed to identify novel direct cell surface receptors of our ligand of interest.
At any time, we experienced great support of Dualsystems Biotech. They kindly helped to find optimal conditions for our purposes and provided help with any kind of question before, during and after the experiment. LRC-TriCEPS allowed us to identify novel cell surface receptors of our ligand, which we could successfully validate in different cell types and with different biochemical assays. The identification of a T cell co-receptor for staphylococcal superantigens had been challenging due to the structural features of the interaction and its kinetics. However, working with Dualstystems Biotech AG, and with Dr. Paul Helbling in particular, and using the LRC-TriCEPS technology, we were able to identify a candidate that was subsequently corroborated by biochemical and functional assays.
