Here you can download an example of a LRC-TriCEPS report where we identified the target of a ligand of interest. In this specific experiment we had three treatment arms, the ligand of interest, a positive control (Transferrin) and a negative control (bovine serum albumin). As there were three treatment arms, three comparisons of the relative abundance of the identified proteins are shown.
During the pretest experiments https://www.dualsystems.com/pretest-triceps-service-kit/ it was evaluated that the expression of the unknown targets are dependent on activation of T-cells, therefore the cells were activated accordingly. Further, by the use of Flow-TriCEPS (https://www.dualsystems.com/pretest-triceps-service-kit/ it was determined that the best way to add the ligands to the cells in this specific case is pH 7.4, for 30 minutes at 22°C. Alternatively, if unknown targets are expressed in strongly adherent cells that are not suited for flow cytometry, binding can also be determined using the Tamra-TriCEPS kit and services https://www.dualsystems.com/tamra-triceps/
In our report, the identified target proteins are further described in more details, showing exactly which peptides of the target proteins were found and their relative abundance in each treatment arm.
