The KickStart system

High level, inducible protein expression in yeast

  • Tight regulation and high expression levels
  • Grow cells in rich medium, such as YPD
  • No medium change required
  • Easy protein purification using the popular Strep-tag

The KickStart protein expression kit has been specifically designed for inducible, high-level expression of recombinant proteins in Saccharomyces cerevisiae.

KickStart expression vectors combine two important features for protein expression:

  • Inducible ADH2 promoter: The ADH2 promoter is induced when glucose becomes depleted in the medium. During early growth in YPD medium glucose levels are high and the ADH2 promoter is inactive - thereby preventing negative effects on yeast growth.
  • Once the cells reach the early stationary phase glucose is depleted from the medium and the ADH2 promoter is induced, producing high amounts of protein.
  • KanMX cassette: KickStart™ vectors feature a KanMX cassette, which allows selection with G418 in rich medium. The use of rich medium such as YPD will significantly enhance cell densities and protein yield.

What are the advantages?

  • High-level, inducible expression
  • No addition of reagents or change of medium needed for induction
  • Tight regulation of induction allows expression of toxic proteins
  • Popular Step-tag for quick and easy protein purification
  • Protease deficient expression strain for higher protein yields
  • Optimized extraction buffers supplied with the kit

Expression of beta-galactosidase from pKS1-ST

pKS1-ST-Gal was transformed into the expression strain DSY-5 and grown on YPD plates supplemented with 60 µg/ml G418. Several colonies were mixed and inoculated into YPD supplemented with 60 µg/ml G418. Aliquots were taken at the indicated time points and used to prepare total cell extracts. Beta-galactosidase was detected by Western blotting against the HA epitope tag. The red arrow indicates the time point where glucose in the medium is depleted.

Beta-galactosidase expressed from pKS1-ST is correctly folded and active

Activity of beta-galactosidase in total extracts was assessed at the indicated time points using the HTX beta-galactosidase assay kit (P01002).
