DUALmembrane services
The DUALmembrane system is a yeast-based screening system to identify interactions involving integral membrane proteins and membrane-associated proteins.
Contrary to the yeast two-hybrid system, the DUALmembrane system tests interactions at the membrane and allows the use of virtaully all types of full-length integral membrane proteins, including G protein-coupled receptors, ion channels and receptor tyrosine kinases.
Applications
Identification of novel interactors for your membrane protein of interest
Testing of pairwise interactions between a set of known membrane proteins
Screening for agents which disrupt protein-protein interactions
Service description
- Determination of appropriate bait vector together with client
- Cloning of cDNA encoding the protein of interest into bait vector
- Functional assay to confirm proper expression and membrane localization of bait
- Pilot screens to adjust selection conditions
- Screening of bait against a cDNA library of your choice
- Analysis of all interactors from the screen
- Removal of false positives by database filtering
- You receive a set of interactors of your membrane protein of interest
- Optional: delivery of plasmids
Key benefits
- Investigate protein interactions involving integral membrane proteins and membrane-associated proteins
- Screen cDNA libraries to identify novel interactors of membrane proteins
- Tested technology, suitable for GPCRs, RTKs, ion channels, etc., see our references
- Complete coverage: all interactors are analyzed
- Normalized cDNA libraries increase coverage and reduce non-specific interactions
