Part I: Bait construction and analysis

Bait cloning
Detection of the bait in total yeast extracts (Western blot)
Self-activation test

We contact you with the results of Part I. Based on the intermediate report you decide whether you want to go ahead with a screen. If you decide to abandon the screen at this step, we will only charge a setup fee.

Part II: Library screen and analysis of interactors

Pilot screen to determine optimal selection conditions
Library screen against cDNA library of your choice
Picking of all primary clones from the screen (up to 192 clones)
Analysis of up to 96 primary clones by prey plasmid isolation and 5'end sequencing*
BLAST search and clone grouping according to confidence level

* Additional clones can be analyzed upon request for a flat fee.

Optional: Specificity test

Bait dependency test to confirm the specificity of the interaction

You receive:

5'end sequences of all prey clones
Plasmid DNA of all prey clones arrayed in 96 well plates
Clone analysis, including BLAST search results and clone grouping according to confidence levels
Complete report
