Dualsystems Biotech is a service provider for custom proteomics for industry and academia. We are a biotech company specialising in elucidating protein-protein interaction in the living cells focussing at the cell membrane. We identify the targets of your extracellular ligands using the ligand-receptor capture LRC-TriCEPS™ technology. During the interaction of your ligand of interest and its unknown targets and off-targets the cells are still alive and the unknown targets are in their natural microenvironment.

Moreover, with Flow-TriCEPS™ (Prestest TriCEPS™) we provide a tool to use in flow cytometry to identify the best binding conditions for your ligand of interest without the need of a detection antibody. Flow-TriCEPS can also be used in functional assays to test if a TriCEPS coupled ligand is functionally active. Flow TriCEPS couples the ligand by using primary amines (N-Term and Lysines) and since it contains a biotin, can be detected by a streptavidin fluorophore conjugate using flow cytometry.

Detection of the binding of your ligand to the cells using TriCEPS V2.0

Identification of the targets using TriCEPS V2.0 in or at the cell membrane of your ligand (peptide, protein, Antibody, ADC or other primary amine containing molecules) requires that the cells used are expressing the unknown targets. TriCEPS V2.0 is the ideal tool to screen cell lines or primary cells to identify the best cells to use later in the LRC-TriCEPS target identification experiments. Dualsystems offers these pretests for cell selection as both service and kit. Needed for the experiment are the ligand of interest and the cell lines/ primary cells that are expected to express the unknown target. There is no need for a ligand specific antibody.

Ligand receptor capture LRC-TriCEPS™ is a conceptually new technology to analyse the protein interaction of your biologics. We can identify the targets of your protein, antibody and peptide at the cell surface. Dualsystems offers the LRC-TriCEPS™ technology as a service for fee model to the industry and academia. The technology enables the unbiased identification of cell surface targets and off-targets on the living cells without genetic manipulation. Ligand-receptor capture technology can be used for many types of orphan ligands, such as peptides, proteins, antibodies, engineered affinity binders and also for viruses.

TriCEPS V2.0 mediated selection of the cells for target identification using Flow Cytometry (FACS).


Now, Dualsystems provides TriCEPS V2.0 for choosing the best cell type for your LRC-TriCEPS experiment to identify the targets of your ligand: peptide, protein, antibody, ADC or other primary amine containing molecules. TriCEPS V2.0 couples to your ligand at the primary amines – for example Lysines and N-terminus – and since TriCEPS contains a biotin it can be detected using flow cytometry by attaching a fluorophore.

TriCEPS V2.0 is a tool to support the selection of the right cells to identify the unknown target receptors. Further, it can be used as a flow cytometric reagent for testing binding of proteins or other primary amines containing molecules to the cell surface.

Pretests are provided as a service by Dualsystems Biotech AG or pretest kits can be purchased to perform the pretests in-house.

Cellular responses to ligands such as peptides, proteins, pharmaceutical drugs or entire pathogens are generally mediated through interactions with proteins expressed at the cell surface. The LRC-TriCEPS kit is designed to directly identify the target proteins of your ligand on the living cells directly using near-physiological conditions.

Dualsystems now offers the pretest TriCEPS experiments as services or as a kit.

The pretest test is designed to screen different cell types and decide which cells to use later for the identification of the unknown targets of your ligand of interest in the LRC-TriCEPS main experiment.

In case functional assays are available TriCEPS coupled ligands can also be tested to see if a similar output is achievable with a TriCEPS coupled ligand compared to a ligand that is not coupled to TriCEPS.

In addition, TriCEPS V2.0 can be utilized as Flow Cytometry (FACS) reagent for detecting binding of primary amine containing molecules to cells.

As first step, the TriCEPS v.2.0 molecule is coupled to the ligand of interest (peptide, protein, Antibody, ADC or other primary amine containing molecules) and to the positive control ligand (e.g. transferrin) and negative control ligand (e.g. glycine). This coupling reaction is tested with Dot blot to assess if the coupling worked.

Then, the TriCEPS coupled ligands are then added to the non-oxidized cells to show that the ligand binds to the unknown targets at the cell surface and TriCEPS does not interfere with the ligand receptor interaction.

Flow Cytometry tests with TriCEPS coupled ligand on oxidized cells can be further performed to test cell viability and ligand target binding as it will occur in the main LRC-TriCEPS experiment.

After successful completion of all pretests the main experiment can be planned and carried out as described in the LRC-TriCEPS user manual choosing one of the service options provided by Dualsystems Biotech AG.
